In Star Wars, clones have always been solely genetic. Aside from clones that have had their memories manually transplanted from another being or clones possessed by the spirit of another being (like Palpatine in both continuities during Dark Empire/TRoS), all clones are completely unique mentally-speaking. Rex, Cody, Sister, Omega, Boba, etc. are all genetically identical (with the exception of Omega being, as far as we know, afab and not transfem), but mentally they are all unique, both in comparison to Jango and from to each other.
If the rules of irl genetics apply to in universe then a female clone is entirely possible.
Sex determination is likely due to a gene, usually found on the Y chromosome called SRY. This gene, when working causes the formation of male reproductive glands (testes).
If the SRY gene fails to express then a person with XY chromosomes can be female.
So Sister may well have Jango's XY chromosomes, making her genotype male, but have the SRY gene work incorrectly during foetal development, giving her a female phenotype (characteristics which actually show irl).
Alternatively, Sister may have a genetic/developmental issue in which her cells are unable to take up testosterone. Thus reducing male characteristics.
You can have all the genes a male has but gene expression determines your real life gender characteristics during foetal development.
Interestingly during the process of meiosis (formation of sex cells) the SRY gene can translocate and attach itself to an X chromosome, meaning you can have the XX female genotype but actually be male.
So yes, you can have entirely different characteristics to those which your basic genetic structure suggests and SW has, in this case, got it right. Not common when it comes to SW and scientific evidence.
Genetics is fascinating but also prone to change. Biology is often called the science of exceptions because of its tendency to produce organisms which 'break the rules'. The truth, however, is that we probably don't understand the rules yet.

@Ctmeh Even with genetics in mind it's explicitly confirmed Sister is trans, mtf. She would have been assigned male at birth (er, test-tube-escape?) by the Kaminoans and transitioned at some point between the time she was "born" and the time we see her in the Queen's trilogy. Given the consistency across nearly all other clones genetically it's a massive stretch to say her genetics are even slightly female. That and calling her a genetic anomaly would be offensive to trans people and her having female traits from a genetic standpoint would diminish her status as trans rep.
Fair enough. I was exploring possible genetic influences on gender/gender choice and the ways in which set genetics (such as a DNA template) can change.
